Identification of Armeniacae Semen Amarum,Persicae Semen and sweet apricot kernel materials based on UPLC characteristic chromatograms
Objective To establish UPLC characteristic chromatogram of 3 Rosaceae medicinal materials to identify and control the quality of Armeniacae Semen Amarum,sweet apricot kernel and Persicae Semen combined with chemometrics analysis.Methods The gradient elution was performed on the Agilent ZORBAX SB-Aq column(2.1 mm×100 mm,1.8 μm)with the mobile phase of acetonitrile-0.2%phosphoric acid solution at 0.3 mL·min-1.The column temperature was 25℃.Totally 35 batches of 3 Rosaceae medicinal materials were collected,the characteristic spectrum was established and the similarity was evaluated.The chemical components of Armeniacae Semen Amarum,sweet apricot kernel and Persicae Semen were studied by cluster analysis and orthogonal partial least squares discriminant analysis.Results The characteristic chromatograms of the 3 seed medicinal materials of Rosaceae were obviously different.The Armeniacae Semen Amarum,sweet apricot kernel and Persicae Semen could be distinguished by peak 2 and 4.Totally 5 characteristic peaks were preliminarily identified through MS identification combined with relevant literature research,and 2 components were further identified as amygdalin and prunasin by comparison with the reference substances.Cluster analysis accurately distinguished 3 types of medicinal materials,with high intra species similarity and low inter species similarity.The results of orthogonal partial least squares discriminant analysis were consistent with those of characteristic chromatogram analysis and cluster analysis,and amygdalin was selected as a Q-marker for the 3 medicinal materials.Conclusion The method is simple,rapid and accurate,which can provide reference for the identification and quality control of 3 Rosaceae medicinal materials.
