Differences in chemical compositions of different basal herbs of epimedium and Wushan epimedium
Objective To determine the differences in chemical compositions of different basal herbs of epimedium and Wushan epimedium by UPLC-Q-TOF-MS,and identify and control the quality epimedium and Wushan epimedium based on their chemical compositions. Methods An Agilent SB-C18(2.1 mm×100 mm,2.7 μm)column was used with 0.1%formic acid water-methanol(+)and water-methanol(-)as the mobile phases for gradient elution at a flow rate of 0.4 mL·min-1.The mass spectrometry was performed with an electrospray ionization(ESI)ion source, and the data were collected in both positive and negative ion modes. The chemical components in different basal epimedium and Wushan epimedium herbs were identified via retention time, relative molecular mass and secondary fragment ions.Based on the identified compounds, the chemical compositions in the different basal epimedium and Wushan epimedium herbs were differentially analyzed by SIMCA software. Results Totally 37 chemical constituents such as epimedin A,epimedin B,epimedin C and icariin were identified in the different basal epimedium and Wushan epimedium herbs. The relative contents of 12 constituents, such as icariin L,2'-O-rhamnosylicariside Ⅱ and magnoflorine in the basal epimedium and Wushan epimedium herbs had obvious differences, and the differential compounds were flavonoids mainly. Conclusion The compounds found in this experiment can be used as the indicators to distinguishing different basal epimedium and Wushan epimedium, laying a foundation for the identification and quality control of the 5 kinds of epimedium herbs.
