Effect of aesculetin on the proliferation,migration,invasion and glycolysis of human gastric cancer SGC-7901 cells
Objective To determine the effect and mechanism of aesculetin(AES)on the proliferation,migration,invasion and glycolysis of human gastric cancer SGC-7901 cells.Methods The cell proliferation was detected by MTT assay.The cell migration and invasion were detected by scratch test and transwell test.Glucose uptake test and lactic acid content test were used to detect the glycolysis of cells.The expression level of migration,invasion and glycolytic related proteins were detected by Western blot.Results AES inhibited the proliferation of SGC-7901 cells in a dose-dependent manner.The migration,invasion and glycolysis of SGC-7901 cells decreased gradually with the gradient increase of AES doses(P<0.05).AES down-regulated the protein expression of HIF-1α,MMP-2,MMP-9,GLUT1 and LDHA in SGC-7901 cells(P<0.05).Conclusion AES can inhibit the proliferation,migration and invasion of human gastric cancer SGC-7901 cells,and reduce the glycolysis level by inhibiting the glucose uptake and lactic acid production of human gastric cancer SGC-7901 cells.AES may inhibit the migration,invasion and aerobic glycolysis of SGC-7901 cells by influencing hypoxia-inducing factor HIF-1α.
