基于HPLC-Q-Exactive-MS技术定性分析冠心七味片的化学成分

目的 采用高效液相色谱-四极杆/静电场轨道阱高分辨质谱(HPLC-Q-Exactive-MS)技术对冠心七味片的主要成分进行快速、全面的定性分析.方法 采用AgilentZORBAX SB-Aq色谱柱(4.6 mm×150 mm,5μm),流动相为甲醇(A)-0.5%甲酸水(B),流速为1.0 mL·min-1,柱温35 ℃,进样量10 μL,质谱采用正、负离子扫描模式,扫描范围m/z 110~1200.基于化合物的精确质荷比、二级质谱裂解碎片,结合自建数据库比对所含成分进行分析鉴定.结果 在冠心七味片中共鉴定出67个化合物,包括30个黄酮类、18个有机酸类、8个萜类、5个木脂素类、4个香豆素类、2个其他类化合物.结论 该HPLC-Q-Exactive-MS方法能够对冠心七味片化学成分进行全面、快速定性分析,为其质量控制、药效物质基础研究提供基础.
Qualitative analysis of chemical constituents of Guanxin Qiwei tablets based on HPLC-Q-Exactive-MS technique
Objective To rapidly and comprehensively determine the main components of Guanxin Qiwei tablets by high performance liquid chromatography-quadrupole/electrostatic field orbit-trap high resolution mass spectrometry(HPLC-Q-Exactive-MS).Methods The chromatography was performed on an Agilent ZORBAX SB-Aq column(4.6 mm×150 mm,5 μm)with mobile phase consisting of methanol(A)-0.5%formic acid water(B)at 1.0 mL·min-1.The column temperature was 35 ℃ and the sample size was 10 μL.Positive and negative ion scanning modes were used for the mass spectrometry.Scanning range of m/z was 110 to 1200.Based on the precise mass charge ratio and secondary mass spectrometry of the compounds,the analysis and identification of the components were conducted with the self-built database.Results Totally 67 compounds,including 30 flavonoids,18 organic acids,8 terpenoids,5 lignans,4 coumarins and 2 other compounds were identified in Guanxin Qiwei tablets.Conclusion The HPLC-Q-Exactive-MS method can rapidly and comprehensively analyze the chemical components in Guanxin Qiwei tablets,which provides reference for the quality control and research on pharmacodynamic substances.
