Effect of flavanomarein on the intestinal function in mice with spleen deficiency diarrhea based on intestinal flora
Objective To explore the effect of flavanomarein on the intestinal function in mice with spleen deficiency diarrhea based on intestinal flora.Methods Mice were randomly divided into a blank group and a modeling group,the modeling group was orally given rhubarb decoction combined with a humid environment method to establish a spleen deficiency diarrhea model.Subsequently,the modeling group was randomly divided into a model group,a flavanomarein group and a positive control group.The blank group and the model groups were given distilled water by gavage,while the treatment groups were given corresponding drugs by gavage.The general physical signs and pathological changes in the colon tissue of mice were observed,the levels of inflammatory factors were measured,the intestinal flora were analyzed with 16S gene sequencing technology,and the levels of short chain fatty acids(SCFA)in mouse feces were detected with 1H-NMR technology.Results Flavanomarein upregulated the abundance of beneficial intestinal flora,the diversity and richness of intestinal flora in mice with spleen deficiency diarrhea,such as Bacteroidota,Muribalactaceae and Eubacterium coprostanogenes group.It also downregulated the abundance of harmful intestinal flora,such as Firmicutes,Proteobacteria,Campilobacterota,Helicobacter,Ruminococcus-torques-group and Colidextribacter,adjusting the abundance of dominant intestinal flora towards that of the blank group.The beneficial intestinal flora were negatively correlated with the serum inter leukin(IL)-6,tumor necrosis factor α(TNF-α),and vasoactive peptide(VIP)in mice with spleen deficiency diarrhea,but positively correlated with IL-10,and gastin(GAS).The opposite was true for the harmful intestinal flora.Conclusion Flavanomarein may regulate the intestinal flora balance in mice with spleen deficiency diarrhea,inhibit the serum and intestinal inflammation,protect and repair intestinal mucosa,enhance immune function,and change intestinal SCFA levels,effectively inhibiting the occurrence and deterioration of spleen deficiency diarrhea in mice.
