UPLC characteristic chromatogram and multi-index component content determination of Folium Mahoniae
Objective To establish the characteristic chromatogram of Folium Mahoniae,and to analyze the quality differences in the medicinal materials from different origins,and evaluate its quality level by the similarity,cluster analysis and principal component factor analysis.Methods The chromatography was performed on Waters CORTECS shield RP 18(100 mm×2.1 mm,1.6μm)column with acetonitrile-0.05 mol·L-1 potassium dihydrogen phosphate solution as the mobile phase.Gradient elution was conducted at a column temperature of 30 ℃ and flow rate of 0.3 mL·min-1.The detection wavelength was 326 nm,and 15 batches of Folium Mahoniae were detected.Results Totally 10 characteristic peaks were identified,and 3 components were identified confirmed as methyl 4-O-caffeoylquinate,methyl chlorogenate,and methyl caffeate.The similarity of 15 batches sample was between 0.913~0.987.The results of cluster analysis showed that 15 batches of medicinal materials from different origins were clustered into 3 categories,and the two main components were extracted,with cumulative variance contribution rate reaching 87.842%.The content of methyl 4-O-caffeoylquinate in multiple batches of medicinal materials was 0.015%~0.038%,methyl chlorogenate content was 0.145%~0.380%,methyl caffeate content was 0.014%~0.166%,and the total content was 0.189%~0.575%.Conclusion The study establishes the characteristic chromatogram and multi-index component content determination method for Folium Mahoniae,which can distinguish the authenticity of Folium Mahoniae and evaluate their quality.
