Fingerprint of Yinhua antiviral oral liquid and multi-index determination content based on UPLC-Q-TOF-MS
Objective To establish a UPLC-Q-TOF-MS fingerprint of Yinhua antiviral oral liquid identify its main chemical compositions,and to determine the content of 6 chemical compositions.Methods UPLC-Q-TOF-MS was used.Under positive and negative ion modes,an Agilent ZORBAX Eclipse Plus C18(3.0 mm×100 mm,1.8 μm)column was used with gradient elution of an aqueous phase(A,containing 0.1%formic acid)and an organic phase acetonitrile(B)as the mobile phase at a flow rate of 0.4 mL·min-1 and an injection volume of 1 μL.Fingerprints of 12 batches of Yinhua antiviral oral liquid were established and the common peaks were determined.Similarity evaluation,cluster analysis,principal component analysis and partial least squares discriminant analysis were performed.The chemical components of common peaks were identify through primary and secondary mass spectrometry,and compared with reference materials.Results Totally 32 peaks were identified in the fingerprint spectra of 12 batches of Yinhua antiviral oral liquid under positive and negative ion modes.The fingerprint similarity was≥0.978.The samples were divided into categories four or three by cluster analysis,which was consistent with principal component analysis.Eight components including wogonside,oroxyloside,forsythoside I,and isochlorogenic acid B were screened out as the hallmark compounds for the differences between various batches of samples.Conclusion The fingerprint combined with multi-index content determination method established in this study is stable,repeatable and specific,which can provide reference for the quality evaluation and clinical application of Yinhua antiviral oral liquid.
