目的 建立芡实配方颗粒高效液相色谱特征图谱,并测定其中腺苷、没食子酸、鸟苷和鞣花酸的含量.方法 以Shim-pack GIST C18-AQ(4.6 mm×150 mm,3.0 μm)为色谱柱,以乙腈-0.5%甲酸溶液为流动相进行梯度洗脱,柱温20℃,流速1.0mL·min-1,检测波长258 nm.建立芡实配方颗粒特征图谱,并借助中药色谱指纹图谱相似度系统评价其相似度.结果 芡实配方颗粒特征图谱共确定了9个共有峰,并指认了7个共有峰成分,其中峰2为腺苷、峰3为没食子酸、峰4为鸟苷、峰6为短叶苏木酚酸、峰7为柯里拉京、峰8为尿石素M5、峰9为鞣花酸.样品相似度均在0.96以上.含量测定结果显示,腺苷含量为0.169~0.344 mg·g-1,没食子酸含量为0.469~1.956 mg·g-1,鸟苷含量为0.162~0.439 mg·g-1鞣花酸含量为0.101~0.617 mg·g-1.结论 建立的芡实配方颗粒特征图谱及主成分含量测定方法重现性好、适用范围广,可为芡实配方颗粒的质量控制提供参考.
HPLC specific chromatogram and quantitation of 4 components in Euryales semen formula granules
Objective To establish the HPLC specific chromatogram of Euryales semen formula granules and to determine the content of adenosine,gallic acid,guanosine and ellagic acid.Methods HPLC analysis of Euryales semen formula granules was conducted on Shim-pack GIST C18-AQ(4.6 mm×150 mm,3.0 μm)column with the mobile phase of acetonitrile-0.5%formic acid system in gradient elution mode at a flow of 1.0 mL·min-1.The ultraviolet detection wavelength was 258 nm and the column temperature was 20 ℃.The similarity was evaluated by the traditional Chinese medicine chromatography fingerprint map similarity system.Results Totally 9 Nine common peaks were identified in the characteristic chromatogram and 7 peaks were comfirmed.Peak 2 was adenosine,peak 3 was gallic acid,peak 4 was guanosine,peak 6 was brevifolin carboxylic acid,peak 7 was corilagin,peak 8 was urolithin M5,and peak 9 was ellagic acid.The similarity of multiple batches of samples was over 0.96.The content of adenosine,gallic acid,guanosine,and ellagic acid ranged 0.169~0.344 mg·g-1,0.469~1.956 mg·g-1,0.162~0.439 mg·g-1,and 0.101~0.617 mg·g-1.Conclusion The characteristic HPLC chromatogram of Euryales semen formula granules is simple repeatable,which can comprehensively and truly reflect the quality difference in Euryales semen formula granules.
