Flavonoids and antioxidant activity of water decoction prepared by Aurantii Fructus Immaturus from different origins
Objective To compare the differences in flavonoid components and antioxidant activity of water decoctions by Aurantii Fructus Immaturus of different origins, and to provide reference for the quality evaluation and clinical use of Aurantii Fructus Immaturus. Methods HPLC method was used to simultaneously determine 8 flavonoid components(narirutin, naringin, hesperidin, neohesperidin, naringenin, hesperetin, nobiletin and tangeretin)in the water decoction of Aurantii Fructus Immaturus. The differential chemical compositions were screened by multivariate statistics. The antioxidant activities of water decoction from different origins of Aurantii Fructus Immaturus were determined by 1,l-diphenyl-2-picrylhydrazyl(DPPH)method and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)diammonium salt(ABTS)method. The correlation between the content of 8 components and antioxidant activity in the water decoction of Aurantii Fructus Immaturus was evaluated by grey correlation and bivariate correlation analysis. Results The main differential components in the water decoction of different origins of Aurantii Fructus Immaturus were naringin and neohesperidin. There was almost no naringin and neohesperidin in the water decoction of Citrus sinensis, while these two components were higher in the water decoction of Citrus aurantium. The antioxidant activity of the water decoction of Citrus aurantium was significantly better than that of Citrus sinensis(P<0.05).Grey correlation and bivariate correlation analysis showed that the core pharmacodynamic components for the antioxidant effect in the water decoction of Citrus aurantium included hesperidin, neohesperidin and nobiletin;while the core pharmacodynamic components for antioxidant effect in the water decoction of Citrus sinensis were narirutin, hesperetin and nobiletin. Conclusion The HPLC method is established for the determination of 8 flavonoids in the water decoction of Aurantii Fructus Immaturus, with differences in the content of flavonoids and antioxidant capacity in water decoctions of different origins of Aurantii Fructus Immaturus.
