Flavonoids from Dendrobium fimbriatum and its protective effect on corticosterone-induced PC 12 cell damage
Objective To determine flavonoid components from Dendrobium fimbriatum and its protective effect on PC 12 cell damage induced by corticosterone.Methods The compounds were separated and purified by polyamide resin,Sephadex LH-20 column chromatography,and silica gel column chromatography.The structures of the isolated compounds were analyzed and identified with nuclear magnetic resonance spectroscopy.The activity of the compounds was tested by in vitro cell assay.Results Totally 10 flavonoids and their glycosides were isolated from Dendrobium fimbriatum,and identified as vicenin-2(1),isoviolanthin(2),violanthin(3),apigenin-6-C-α-L-rhamnosyl-8-C-β-D-quinovoside(4),apigenin-6-C-α-L-arabinosyl-8-C-β-D-arabinoside(5),apigenin-6,8-di-C-α-L-arabinoside(6),chryseriol-7-O-β-D-glucoside(7),apigenin-6-C-α-L-rhamnoside(8),isovitexin(9),and apigenin(10).Compounds 3~6 and 8~10 significantly improved the survival rate of PC 12 cell damage induced by corticosterone.Conclusion Compounds 4,5,and 7~10 have been isolated from Dendrobium fimbriatum for the first time.Compounds 5,7 and 8 have been isolated from Dendrobium for the first time.Compounds 3~6 and 8~10 show neuroprotective effect on PC 12 cell damage induced by corticosterone.
