高效液相色谱法测定α-硫辛酸绿茶片中α-硫辛酸的含量

摘要
目的 建立α-硫辛酸绿茶片中α-硫辛酸的含量测定方法.方法 利用高效液相色谱(HPLC)技术,采用Alltima C18柱(250 mm×4.6 mm,5 μm),流动相为乙腈-0.1%磷酸溶液(55∶45),流速为1.0 mL·min-1,检测波长为215 nm,柱温为30 ℃.结果 α-硫辛酸在102.6~307.7 μg·mL-1与峰面积呈良好的线性关系,相关系数(r)为1.000;精密度、重复性和稳定性考察RSD均小于2.0%;3个浓度的加样回收率为98.44%~100.2%,RSD值均小于1.0%.结论 该方法准确、简单、快速,能够用于α-硫辛酸绿茶片中α-硫辛酸的含量测定.
Abstract
Objective To establish a method for the determination of α-lipoic acid in α-lipoic acid with green tea tablets.Methods HPLC was carried out,and the experiment was conducted with an Alltima C18 column(250 mm × 4.6 mm,5 μm).The mobile phase was acetonitrile-0.1%phosphoric acid solution(55∶45)at the flow rate of 1.0 mL·min-1.The detection wavelength was 215 nm and the column temperature was 30 ℃.Results α-lipoic acid showed a good linearity with the peak area at 102.6~307.7 μg·mL-1,and the correlation coefficient(r)was 1.000.The RSD of precision,repeatability and stability were less than 2.0%.The average recovery of 3 concentrations was 98.44%~100.2%,and the relative standard deviation was less than 1.0%.Conclusion The method is fast,simple and accurate,and can be used to determine α-lipoic acid in α-lipoic acid with green tea tablets.
