Identification and traceability of microbial pollution in traditional Chinese medicine tablets and its risk evaluation
Objective To trace and analyze the source of contamination of bile salt resistant gram-negative bacteria in the powder of traditional Chinese medicine tablets through RiboPrinter method.Methods Totally 16 strains of bile salt resistant gram-negative bacteria isolated from the powder of traditional Chinese medicine tablets and the environment were identified and compared with VITEK2 Compact,RiboPrinter,16SrRNA,and MALDI-TOF MS methods.Finally,the RiboPrinter method was used to trace the gene fingerprints of the strains with consistent identification results.Results The identification of the 6 strains from the powder of traditional Chinese medicine tablets was consistent with 1 strain from the boiling dryer cloth bag,namely Pantoea calida.Through comparison,MALDI-TOF MS had a faster identification speed and higher separation rate.RiboPrinter traceability analysis showed that the 7 strains of Pantoea calida had extremely high homology,with a similarity of 0.94±0.03.Conclusion Based on the RiboPrinter traceability and BioNumerics analysis,the contamination of the the powder of traditional Chinese medicine tablets may come from the boiling dryer bag.Corresponding measures should be taken to solve the problem.
