Effect of Naoluoxintong on anti-lipid peroxidation injury and mitochondrial damage after ischemic stroke
Objective To determine the protective effect of Naoluoxintong(NLXT)on lipid peroxidation injury and mitochondrial damage after ischemic stroke. Methods SPF male SD rats were used to establish the cerebral ischemic/reperfusion model by suture method. The neurological function was evaluated according to Zea Longa standards. The model animals with scores of 2~3 were randomly divided into a model group, and NLXT low, middle, and high dose groups.A sham operation group and a normal group were also established. The rats were treated with NLXT for 7 days, meanwhile, rats in the normal and the sham-operated groups were gavaged with saline. The effect of NLXT on living condition, body weight, neurological function score, brain histopathology, lipid peroxide(LPO)content,4-hydroxyenal(4-HNE)level,mitochondrial morphology and hypoxia inducible factor-1α expression were observed. Results Compared with the model group,NLXT improved the rat's quality of life, increased the body weight and their neurological function, reduced the accumulation of LPO,4-HNE and HIF-1α,and protected against the pathological damage of the brain tissue and the structure of mitochondria. Conclusion NLXT plays a neuroprotective role by inhibiting lipid peroxidation and protecting mitochondrial damage after ischemic stroke.
