目的 基于UHPLC-Q-Exactive Orbitrap MS技术对静宁方给药后的大鼠入脑、入肠及粪便中的成分进行分析和鉴定.方法 采用UHPLC-Q-Exactive Orbitrap MS,以 0.1%甲酸-乙腈(A)和0.1%甲酸-水(B)为流动相进行梯度洗脱,流速0.2 mL·min-1,进样量5 μL,加热电喷雾离子化源(HESI),正、负离子扫描.结合对照品比对、文献报道、数据库比对等方式对成分进行分析鉴定.结果 共鉴定出 42 种成分,其中脑、回肠、结肠、粪便中分别鉴定出9、33、20、27种成分.结论 本实验初步阐明静宁方入脑、入肠及粪便成分,为阐明其药效物质基础及作用机制提供了依据.
Brain,intestinal and feces components of Jingning formula based on UHPLC-Q-Exactive Orbitrap MS technology
Objective To identify and analyze the components in the brain,intestine and feces of rats after the administration of Jingning formula by UHPLC-Q-Exactive Orbitrap MS technology.Methods UHPLC-Q-Exactive Orbitrap MS was used.The mobile phase was 0.1%formic acid-acetonitrile(A)and 0.1%formic acid-water(B)with gradient elution,the flow rate was 0.2 mL·min-1,and the injection volume was 5 μL.The mass spectrometry conditions were included both positive and negative ion scannings,and the ion source was heated with electrospray ionization.The chemical compositions were analyzed by comparison with reference materials,literature reports and databases.Results Totally 42 components were identified,of which totally 9,33,20,and 27 components were identified in the brain,ileum,colon and feces,respectively.Conclusion The compositions of Jingning formula into the brain,intestine and feces are elucidated,which provides a basis for clarifying the material basis and action mechanism of Jingning formula.
