Orexin is an excitatory neuropeptide secreted by the hypothalamus,with many physiological effects,including regulating food intake and energy balance,sleep-wake state,addiction,anxiety and depression,etc.At present,only the complete study of orexin receptor antagonists for insomnia has been reported and made into drugs.Compared with orexin receptor antagonists,research on orexin receptor agonists is still very few,despite its prospect in development.This article reviewed the research on orexin receptor agonists,their potential roles in the treatment of narcoleptic disease,reversal of drug-induced respiratory depression,and promotion of cancer cell apoptosis.This article also reviewed the research in recent years and discussed the existing problems to provide theoretical basis and ideas for the development of new drugs.
