Steroid alkaloids in Veratrum mengtzeanum Loes.and their anti-inflammation activity
Objective To determine the steroid alkaloids and their anti-inflammation activity in Veratrum mengtzeanum Loes.,a medicinal herb in Yunnan.Methods Column chromatography,reverse phase chromatography,gel chromatography and other separation methods were used to separate and purify the ethanol extract from the roots of Veratrum mengtzeanum Loes..The chemical structures of these alkaloids were determined by spectroscopic data and physical and chemical properties,and their anti-inflammation activity was measured.Results Totally 8 steroid alkaloids,including 3-angeloylzygadenine(1),3-veratroylgemine(2),jervine(3),verazine(4),vermitaline(5),3-angeloylzygadenine-β-N-oxide(6),cyclopamine(7),and veratramine(8),were isolated and identified from the roots of Veratrum mengtzeanum Loes.Compounds 2,4,and 7 were extracted from this plant for the first time.Notably,compound 8 exhibited obvious inhibition on the production of nitric oxide induced by lipopolysaccharide.At 50 μmol·L-1,the inhibition rate of nitric oxide production was(47.01±0.41)%.Moreover,it effectively reduced the protein expression levels of inflammation factors COX-2 and iNOS in a dose-dependent manner.Conclusion This study provides substance basis for the anti-inflammation and utilization of Veratrum mengtzeanum Loes..
