Pharmacokinetics of 4 components of Shensong Yangxin capsules in rat plasma
Objective To determine the pharmacokinetics of palmatine,berberine,schisandrin A,and tanshinone ⅡA in rat plasma of Shensong Yangxin capsules.Methods Rats were orally administered with a dosage of 0.43 g·kg-1 Shensong Yangxin capsules,and blood samples were collected at different time points.Carbamazepine was used as the internal standard.LC-MS/MS method was established to determine the blood drug concentrations of palmatine,berberine,schisandrin A,and tanshinone ⅡA in rat plasma and calculate the main pharmacokinetic parameters.Results Palmatine,berberine,schisandrin A,and tanshinone ⅡA showed good linear relationship within their respective ranges.The precision and accuracy,extraction recovery rate,and matrix effect all met the requirements for biological sample determination.The pharmacokinetic parameters tmax,Cmax,t1/2,AUC0~t and AUC0~inf of the above 4 components were calculated based on PKSolve.Conclusion The established LC-MS/MS analysis method is sensitive,quick,and accurate,and can be applied to the main active ingredients of Shensong Yangxin capsules in rat plasma,providing a reference for research and rational use of Shensong Yangxin capsules.
