Effect of astaxanthin on myocardial fibrosis in the rat model of myocardial infarction
Objective To determine the effect of astaxanthin on myocardial fibrosis after the myocardial infarction(MI)and its possible mechanism.Methods The MI model was establshed in the astaxanthin(ASX)group and the MI group rats by ligation of the coronary artery.In the sham-operation(Sham)group pericardiotomy was performed,but not ligation.The rats were dosed with astaxanthin[the ASX group,100 mg/(kg·d)]or distilled water(the Sham group and the MI group)by lavaging daily 1 day before the operation.Myocardial fibrosis was observed by Masson staining 28 days post-operatively.Type Ⅰ collagen matrix metalloproteinase-9(MMP-9)and p38MAPK protein expression were detected at the infarcted border zone by Western blot.Results Compared with the MI group,astaxanthin reduced the collagen volume fraction,decreased the expression of MMP-9,type Ⅰ collagen and phosphorylated p38(P-p38).Conclusion Astaxanthin may affect myocardial fibrosis post MI in rats,likely through the inhibition of phosphorylated p3 8.
