高效液相色谱法测定金莲花软胶囊中有效成分的含量

摘要
目的 建立高效液相色谱法同时测定金莲花软胶囊中荭草苷和牡荆苷的含量.方法 采用乙腈-0.1％磷酸溶液通过RD-C18(4.6 mm×250 mm,5 μm)色谱柱进行梯度洗脱,流速1.0 mL·min-1,检测波长340 nm,柱温40℃,进样量10 μL.结果 荭草苷、牡荆苷分别在质量浓度 1.94～194.8 μg·mL-1(r=1.000)和 1.03～103.7 μg·mL-1(r=1.000)内与峰面积呈良好的线性关系;平均加样回收率分别为103.6％和100.2％,RSD分别为0.64％和0.78％(n=6),14批样品荭草苷和牡荆苷的含量分别在3.090～4.729 mg·g-1和1.104～1.402 mg·g-1.结论 所建立的方法简单、准确,可用于金莲花软胶囊的质量控制.
Abstract
Objective To simultaneously determine the content of orientin and vitexin in Jinlianhua soft capsules by HPLC.Methods An RD-C18(4.6 mm ×250 mm,5 μm)chromatographic column was used with a gradient elution of acetonitrile and 0.1％phosphoric acid solution as the mobile phase.The flow rate of 1.0 mL·min-1,the wavelength was 340 nm,with the column temperature of 40 ℃,and the injection volume of 10 μL.Results The linear range of orientin and vitexin was 1.94～194.8μg·mL-1(r=1.000)and 1.03～103.7 μg·mL-1(r=1.000).The average recovery was 103.6％and 100.2％,and RSD was 0.64％and 0.78％(n=6).The contents of orientin and vitexin in the 14 batches ranged 3.090～4.729 mg·g-1 and 1.104～1.402 mg·g-1.Conclusion The method is simple and accuracy,which can be used for the quality control of Jinlianhua soft capsules.
