Correlation of characteristic spectrum and quantitative value transfer analysis of Scolopendra and its decoction pieces,standard decoction and formula granules
Objective To establish the high performance liquid chromatography(HPLC)for the characteristics of Scolopendra and its decoction pieces,standard decoction and formula granules,and to determine the content of hypoxanthine and xanthine at the same time,and the transfer and difference in chemical components among the four.Methods Chromatography was performed on a Dikma Platisil ODS-C18 column(250 mm × 4.6 mm,5 μm).The methanol(A)and water(B)was used as the mobile phase with gradient elution.The flow rate was 1 mL·min-1,and the column temperature was 25 ℃.With the content,common peak transfer number and similarity of characteristic map as the main evaluation indexes,the quantitative value transfer was analyzed.Results There were 8 common characteristic peaks in the characteristic maps of 18 batches of Scolopendra and its decoction pieces,standard decoction and 3 batches of formula granules.The similarity between them and the control characteristic maps was greater than 0.90.Totally 8 components(uracil,tyrosine,hypoxanthine,xanthine,phenylalanine,inosine,guanosine,and tryptophan)were identified and their content was determined without discrete data.Conclusion The main chemical components in Scolopendra and its decoction pieces,standard decoction and formula granules are basically the same.The correlation of HPLC characteristic map is good,and the linearity of the 8 components is good within the determination concentration.The quality evaluation model can reflect the overall appearance of the four forms and components of Scolopendra,and provide data support for the quality control of the formula granules of Scolopendra.
