目的 分析二陈汤化裁方破壁饮片水溶液的化学成分及入血成分.方法 采用超高效液相色谱-四极杆-静电场轨道阱高分辨质谱(UHPLC-Q-Exactive-MS)法对比复方样品、空白血清、含药血清色谱图,分析二陈汤化裁方破壁饮片水溶液在大鼠血清中的原形及代谢产物.结果 从复方样品中共鉴定出151个化学成分,通过对照品比对,最终确定了 11个成分(柠檬酸、绿原酸、橙皮苷、茯苓酸B、熊果酸、天麻素、巴利森苷A、牡荆素、金丝桃苷、人参皂苷Rg1、人参皂苷Rd).共鉴定出33个入血成分,其中28个为原形成分,5个为代谢产物.结论 本实验通过UHPLC-Q-Exactive-MS分析二陈汤化裁方破壁饮片的入血成分,为进一步阐明二陈汤化裁方破壁饮片的药效物质基础提供了参考.
Chemical composition and blood compontents of the water solution of ultrafine granular powder of Erchen decoction modified formula
Objective To determine the chemical compositions and blood compontents in the water solution of ultrafine granular powder of Erchen decoction modified formula.Methods Ultra performance liquid chromatography quadrupole electrostatic field Orbitrap high resolution mass spectrometry(UHPLC-Q-Exactive-MS)was used to compare the chromatograms of compound samples,blank serum and drug containing serum,and analyze the prototype and metabolites of the water solution of ultrafine granular powder of Erchen decoction modified formula in the rat serum.Results Totally 151 chemical components were identified from the water solution of ultrafine granular powder of Erchen decoction modified formula.After comparison with standard substance,11 components(citric acid,chlorogenic acid,hesperidin,poric acid B,ursolic acid,gastrodin,parishin A,vitexin,hyperoside,hinsenoside Rg1,and ginsenoside Rd)were ultimately determined.Totally 33 blood components were identified,28 of which were prototypes and 5 were metabolites.Conclusion The blood components in the water solution of ultrafine granular powder of Erchen decoction modified formula are determined by UHPLC-Q-Exactive-MS,providing a reference for further elucidating the pharmacological substance basis and mechanism of its action.
