不同产地白术挥发性成分HS-SPME/GC-MS分析

目的 分析不同产地白术挥发性成分差异,为白术产地溯源、加工和品质评价提供科学依据.方法 采用顶空固相微萃取技术(HS-SPME)结合气相质谱联用技术(GC-MS),联合层次聚类分析(HCA)、主成分分析(PCA)和偏最小二乘判别分析(OPLS-DA)法对不同产地11批白术挥发性成分进行差异分析.结果 从白术样品中共鉴定出30种挥发性成分,其中共有成分17种,占总挥发性成分含量的88.31%以上;不同产地白术8种挥发性成分含量存在显著差异(VIP>1,P<0.05),其中包含4种品质挥发性成分;浙江和河北白术特征品质挥发性成分为苍术酮,湖南白术特征品质挥发性成分为莪术烯、α-桉叶醇和大根香叶烯B,安徽和重庆白术特征品质挥发性成分为α-桉叶醇.结论 不同产地白术特征品质挥发性成分可为产地溯源、加工和品质评价奠定基础.
Volatile compounds of Atractylodes macrocephala from different regions by HS-SPME/GC-MS
Objective To determine the differences in the volatile compounds of Atractylodes macrocephala from different regions,and to provide reference for the region traceability processing,and quality evaluation of Atractylodes macrocephala.Methods Headspace solid-phase microextraction(HS-SPME),gas chromatography-mass spectrometry(GC-MS),coupled with hierarchical cluster analysis(HCA),principal component analysis(PCA),and orthogonal partial least squares-discriminant analysis(OPLS-DA)were used to analyze the differences in volatile compounds in 11 batches of Atractylodis macrocephala from different regions.Results Totally 30 volatile compounds were identified,17 of which were shared compounds,accounting for more than 88.31%of the total volatile compound content.There were significant differences in the content of 8 volatile compounds in Atractylodes macrocephala from different regions(VIP>1,P<0.05),4 of which contained quality volatile compounds.The characteristic quality volatile compounds of Atractyylodes macrocephala from Zhejiang and Hebei included atractylenol,that from Hunan was curzerene,α-eudesmol,and germacrene B,and that from Anhui and Chongqing was α-eudesmol.Conclusion The volatile compounds of Atractyylodes macrocephala from different regions can be used for region tracing,processing and quality evaluation.
