橘红药材、饮片、标准汤剂与配方颗粒的指纹图谱相关性研究

摘要
目的 建立橘红药材、饮片、标准汤剂、配方颗粒的HPLC指纹图谱,并分析其相关性和差异性.方法 采用CORTECS T3色谱柱(150 mm×4.6 mm,2.7 μm),乙腈(A)和0.05%磷酸水溶液(B)为流动相,进行梯度洗脱,流速为1.1 mL·min-1,检测波长为300nm,柱温为30℃,进样量为10μL.以橙皮苷为参照峰,测定15批橘红药材、饮片、标准汤剂与3批配方颗粒;采用中药色谱指纹图谱相似度评价系统(2012版)进行相似度评价,确定共有峰,并进行聚类分析.结果 橘红药材、饮片、标准汤剂、配方颗粒的HPLC指纹图谱均确定了 12个共有峰,并指认了其中7个峰,峰4为维采宁-2,峰6为阿魏酸,峰7为柚皮芸香苷,峰8为橙皮苷,峰10为川陈皮素,峰11为3,5,6,7,8,3',4'-七甲氧基黄酮,峰12为橘皮素.橘红药材、饮片、标准汤剂、配方颗粒四者与相应对照指纹图谱的相似度均大于0.980;相关性分析的结果表明大部分共有峰显著相关;系统聚类分析结果表明,药材、饮片、标准汤剂、配方颗粒之间无显著性差异.饮片到配方颗粒的质量分数转移率在21.7%~26.8%,均值为24.8%,在均值的±30%范围,未出现离散,说明传递性较好.结论 橘红药材、饮片、标准汤剂、配方颗粒的主要化学成分组成基本相同,指纹图谱相关性良好,可为橘红配方颗粒生产过程的质量控制提供参考.
Abstract
Objective To establish the HPLC fingerprints of Citri Eoxcarpium Rubrum medicinal materials,decoction pieces,standard decoction and formula granules,and analyze their correlation and difference.Methods Gradient elution was performed on a CORTECS T3(150 mm × 4.6 mm,2.7μm)column with acetonitrile(A)and 0.05%phosphoric acid(B)as the mobile phase.The flow rate was 1.1 mL·min-1,the detection wavelength was 300 nm,the column temperature was 30 ℃,and the injection volume was 10 μL.The HPLC fingerprints of 15 batches of Citri Eoxcarpium Rubrum medicinal materials,decoction pieces,standard decoction and 3 batches of formula granules were determined with the hesperidin peak as the reference peak.The Similarity Evaluation System for Chromatographic Fingerprints of Traditional Chinese Medicine(Version 2012)was used to evaluate the similarity and identify common peaks,and the cluster analysis was conducted.Results There were 12 common peaks in the HPLC fingerprints of Citri Eoxcarpium Rubrum medicinal materials,decoction pieces,standard decoction and 3 batches of formula granules,and 7 of them were identified.Peak 4 was vicenin-2,peak 6 was ferulic acid,peak 7 was narirutin,peak 8 was hesperidin,peak 10 was nobiletin,peak 11 was 3,5,6,7,8,3',4'-heptamethoxyflavone,and peak 12 was tangeretin.The similarity between each of the Citri Eoxcarpium Rubrum medicinal materials,decoction pieces,standard decoction and formula granules fingerprints and its corresponding control fingerprint was greater than 0.980.The correlation analysis showed that most common peaks were significantly correlated and the cluster analysis showed no significant difference among the medicinal materials,decoction pieces,standard decoction and formula granules.The content transfer rate from decoction pieces to formula granules was 21.7%~26.8%with an average value of 24.8%.No dispersion was found within±30%of the mean,indicating good transferability.Conclusion The main chemical components of the medicinal materials,decoction pieces,standard decoction and formula granules of Citri Eoxcarpium Rubrum are basically the same.With good correlation,the fingerprint could provide a reference for the quality control of the production of the formula granules of Citri Eoxcarpium Rubrum.
