Exploring the scientific connotation of the theory of"Reinforcing Yang from Yin"in Jinkui Shenqi pills based on fluorescence quenching spectroscopy
Objective To determine the interaction of Jinkui Shenqi pills and their disassembled formulas with bovine serum albumin(BSA)with fluorescence quenching spectroscopy.Methods According to the fluorescence quenching effect of Jinkui Shenqi pills and their disassembled formulas on BSA at 304 K,307 K,and 313 K temperature,Stern-Volmer equation and double log equation were used to analyze the fluorescence quenching type and binding constant of Jinkui Shenqi pills and their disassembled formulas with BSA.Results With the increase of the concentration of quantitative index components among Jinkui Shenqi pills and their disassembled foumulas,the fluorescence intensity of BSA decreased gradually.At the same temperature,the binding constant(the Ka value),of the interaction of the Yin-Yang double supplementing group of Jinkui Shenqi pills with BSA was higher than that of Yang-supplementing group or the Yin-supplementing group with the same quantitative index components;the Ka value of the interaction of the group containing purgatives(poria cocos,rhizoma alismatis,and cortex moutan)with BSA was higher than that of the group without purgatives.Conclusion Jinkui Shenqi pills and their disassembled formulas can combine BSA and quench the fluorescence of BSA,with static quenching as the main type.The interaction of Yin-Yang double supplementing group of Jinkui Shenqi pills with BSA is stronger than that of Yang-supplementing group or Yin-supplementing group with the same quantitative index components,and addition of"three purgatives"further enhances this interaction.
