基于UPLC-Q-Orbitrap-HRMS技术的葛根-丹参药对化学成分分析

目的 采用超高效液相色谱-四极杆-静电场轨道阱高分辨质谱(UPLC-Q-Orbitrap-HRMS)技术分别对以水和50%甲醇溶液为溶剂提取的葛根-丹参药对样品进行化学成分分析.方法 采用 Thermo Scientific Hypersil GOLD C18 色谱柱(150 mm×2.1 mm,1.9 μm),以 0.1%甲酸水-0.1%甲酸乙腈溶液为流动相进行梯度洗脱,流速为0.3 mL·min-1,柱温为30℃.采用加热电喷雾离子源(H-ESI),正负离子检测模式.结果 从水提和50%甲醇醇提样品溶液中均鉴定出104个化合物,其中103个化合物为共有成分,各有1个差异性成分,分别为水提样品中的刺槐苷与50%甲醇醇提样品中的降丹参酮(或其同分异构体).共鉴定出的105个化合物,包括黄酮类成分36个、苯丙素类成分20个、萜类成分24个和其他类成分25个.结论 本研究首次采用UPLC-Q-Orbitrap-HRMS技术对葛根-丹参药对的主要成分进行鉴定,可为葛根-丹参药对的药效成分研究提供科学依据.
Chemical components of Gegen-Danshen herb pair by UPLC-Q-Orbitrap-HRMS
Objective To analyze the chemical components of Gegen-Danshen herb pair extracted with water and 50%methanol solution by UPLC-Q-Orbitrap-HRMS.Methods Thermo Scientific Hypersil GOLD C18 column(150 mm × 2.1 mm,1.9 μm)was used for gradient elution with 0.1%formic acid water-0.1%formic acid acetonitrile solution as the mobile phase.The flow rate was 0.3 mL·min-1 and the column temperature was 30 ℃.A heated electrospray ionization(H-ESI)source was used for HRMS analysis in both positive and negative ion modes.Results In both water and 50%methanol extract samples,104 compounds were tentatively identified:103 common components,and one differentials component,namely acacetin in the water extract and nortanshinone(or its isomers)in 50%methanol extract.Among the identified 105 compounds,there were 36 flavonoids,20 phenylpropanoids,24 terpenoids and 25 other components.Conclusion The main components of Gegen-Danshen herb pair are characterized by UPLC-Q-Orbitrap-HRMS for the first time,which provides reference for the discovery of bioactive components in Gegen-Danshen herb pair.
