Quality standard for Tibetan medicine Rubia wallichiana
Objective To establish the quality standard for Rubia wallichiana.Methods We qualitatively identified Rubia wallichiiana by characters,microscopic identification and thin layer chromatogram.The water content,total ash content and extract content were determined by the methods in Chinese Pharmacopoeia(2020 Edition),and the content of methyl isoalizarin was determined by HPLC.Results Under the microscope,the cross-section showed cork layer,cortex,phloem and xylem,and the powder showed cork cells,parenchyma cells,calcium oxalate needle crystals and ribbed pores.TLC characteristic spot separation was clear.The average content of moisture,total ash and extract in 10 batches of samples were 6.3％,8.5％and 17.5％.At 0.619～30.9500 μg,the linearity of methylisorubidin was good(r=0.9996).The average recovery was 98.3％,RSD was 0.98％,and the average content was 0.0546％.Conclusion The preliminary quality standard for Rubia wallichiana includes moisture content no more than 10.0％,total ash content no more than 12.0％,and the extract no less than 12.0％by hot leaching method with 40％ethanol as the solvent.The content of methylisorubidin shall not be less than 0.030％in the dried product.
