怀山药皮花青素理化特性研究

目的 提取怀山药皮花青素,并测定其理化特性.方法 利用酸性甲醇提取怀山药皮花青素,采用大孔树脂柱层析纯化花青素样品.测定不同pH下花青素的颜色、紫外吸收光谱变化,温度对花青素保存率的影响以及金属离子对花青素稳定性的影响.结果 pH值对怀山药皮花青素稳定性影响较大,随着pH升高,颜色逐渐变淡,稳定性变差;高温对花青素稳定性有一定的影响,低温条件下比较稳定,更容易储存;Fe2+、Fe3+对花青素稳定性的影响较大,而Cu2+、Ca2+、Zn2+和Mn2+等对花青素稳定性无明显影响.结论 怀山药皮花青素稳定性在一定程度上受酸碱度、温度、金属离子的影响.
Physicochemical properties of anthocyanin from the peels of Dioscorea oppostita
Objective To extract anthocyanin from the peels of Dioscorea oppostita(D.opposita)and determine its physicochemical properties.Methods Anthocyanin was extracted by acidic methanol and purified by macroporous resin column chromatography.The color changes and UV absorption spectra of anthocyanin at different pH values,the effect of temperature on the retention rate of anthocyanin,and the effect of metal ions on the stability of anthocyanin were all determined.Results The stability of anthocyanin from the peels of D.opposita was affected by pH obviously.With the increase of pH,the color of anthocyanin turned pale,and anthocyanin became unstable.Anthocyanin was more stable and easier to be stored at low temperature.Fe2+and Fe3+had great effect on the stability of anthocyanin,however,Cu2+,Ca2+,Zn2+and Mn2+had no significant effects on the stability of anthocyanins.Conclusion The stability of anthocyanin from the peels of D.opposita is affected by pH,temperature and metal ions to some extent.
