Objective To determine the inhibition effect of Peptaibol peptide Trichorzin F on colorectal cancer cell SW480 and related mechanism.Methods SW480 cells were cultured in vitro,and MTT assay was used to determine the effect of Trichorzin F on the cell viability.Transwell cell assay was used to detect the effect of SW480 cell invasion ability.Flow cytometry and PI staining were used to investigate the effect of Trichorzin F on the cell cycle distribution and apoptosis.RT-qPCR assay was performed to detect the expression of CCND1,and CDK4 mRNA related to cell cycle regulation.Western blot was used to detect the protein expressions of Bax,Bcl-2 and Cyclin D1,and analyze their effect on Erk1/2 pathway.Results Compared with the blank control group,Trichorzin F inhibited SW480 cell viability and invasion ability.PI staining and flow cytometry showed that Trichorzin F induced the apoptosis of SW480 cells in a time and dose dependent manner,and blocked the cell cycle in the G0/G1 phase.RT-qPCR displayed that Trichorzin F down-regulated the expression of CCND1 and CDK4 mRNA.Western blot showed that Trichorzin F up-regulated the expression of Bax protein,down-regulated the expression of CyclinD1,Bcl-2 protein and the phosphorylation level of Erk1/2.Conclusion Trichorzin F can inhibit the viability and invasion ability of SW480 cells,induce the cell apoptosis,and block the cell cycle process,the mechanism may be related to the regulation of Erk1/2 signaling pathway.
