Effect of aloe emodin on the aggregation and cytotoxicity of Aβ42 in the presence of Cu2+
Objective To determine the inhibitory effect of aloe emodin(AE)on the aggregation and cytotoxicity of Aβ42 in the presence of Cu2+in vitro.Methods An Aβ42 group,a(Aβ42+Cu2+)group,a(Aβ42+AE)group,and a(Aβ42+Cu2++AE)group were set up in the experiment.The fibrotic aggregation process,total aggregation degree,aggregates morphology and cytotoxicity of Aβ42 in each group were studied by thioflavin T fluorescence test,turbidity test,transmission electron microscopy test,and CCK8 cytotoxicity test.Results Compared with the blank group,Cu2+enhanced the total degree of Aβ42 aggregation,induced the Aβ42 aggregation to form irregular aggregates with low β-sheet structure,and greatly reduced the viability of human neuroblastoma cells SH-SY5Y.AE inhibited not only auto aggregation of Aβ42,but also Cu2+-induced aggregation of Aβ42.Compared with the(Aβ42+Cu2+)group,AE reduced the total aggregation degree of Aβ42,induced Aβ42 to form near-round or short rod-like aggregates with low β-sheet structure,and increased the cell viability.Conclusion AE can inhibit Aβ42 aggregation and cytotoxicity in the presence of Cu2+.
