HPLC法测定藏药细果角茴香中原阿片碱含量及其与海拔等影响因素的相关性分析

目的 建立HPLC法测定31批不同产地、不同海拔藏药材细果角茴香中原阿片碱含量.方法 色谱柱 ZORBAX SB-C18(4.6mm×250mm,5μm),流动相为甲醇-0.1 mol·L-1 醋酸铵溶液体系(32∶68,V/V),进样量20 μL,流速0.8 mL·min-1,柱温35 ℃,检测波长289 nm.结果 31批不同产地细果角茴香中原阿片碱含量存在较大差异,处于中高海拔地区的药材中原阿片碱含量偏高,处于高海拔地区的药材中原阿片碱含量偏低.结论 该方法科学、简便、准确,可用于原阿片碱含量测定.不同海拔细果角茴香中原阿片碱含量的差异,对该药材种植、开发和利用具有一定的参照价值.
HPLC determination of the content of protopine in Hypecoum leptocarpum and its influence factors at different altitudes
Objective To establish an HPLC method to determine the content of protopine in 31 batches of Xizang medicine Hypecoum leptocarpum from different origins and altitudes.Methods A ZORBAX SB-C18 column(4.6 mm × 250 mm,5 μm)was used with a mobile phase of methanol-0.1 mol·L-1 ammonium acetate solution(32∶68,V/V).The injection volume was 20 μL,the flow rate was 0.8 mL·min-1,the column temperature was 35 ℃,and the detection wavelength was 289 nm.Results There was a significant difference in the protopine content in Hypecoum leptocarpum from different origins,with higher protopine content in middle to high altitudes,but low levels in high altitudes.Conclusion The method can be used for the determination of protopine content.The difference in protopine content at different altitudes provides reference for the cultivation,development,and utilization of the medicinal herb.
