The aim of this experiment was to develop a high performance liquid chromatographic method for the determination of 5-hydroxymethylfurfural(5-HMF)in different origins of Radix Codonopsis Pilosulae,Radix Polygonui Multiflori,Radix Rehmanniae Preparata and five batches of functional sugars.The separation was performed on a C18(4.6 mm×250.0 mm,5 µm)column with a mobile phase of methanol-water(8∶92)at flow rate of 1.0 mL/min,column temperature of 30℃,sample volume of 10 µL and UV detector at 280 nm.The mean RSD of the peak area was 1.438%in the precision test;the reproducibility test showed good repeatability by HPLC;the RSD of the peak area ranged from 1.338%to 7.307%in the stability test;the spiked recovery showed good results.5-HMF content determination showed that Radix Rehmanniae Preparata had the higher 5-HMF content,while Radix Codonopsis Pilosulae had the lower 5-HMF content,among which Radix Rehmanniae Preparata from Hebei had the highest 5-HMF content.There was no significant difference in the 5-HMF content between different batches of functional sugars,but its content was 90-100 times higher than that of Radix Rehmanniae Preparata from Hebei.Through modern technology,functional sugars has successfully replicated the active ingredient 5-HMF in traditional Chinese medicine,greatly reducing the cost of its application in the breeding industry,which has good development prospects.
