Study on qualitative quality standard of stems and leaves of Millettia speciosa Champ.in Guangdong Province
The objective was to establish the qualitative and quality standard of the stems and leaves of Millettia speciosa Champ.in Guangdong Province.The stems and leaves were collected from 5 regions of Guangdong Province,and the powder was identified by TLC and microstructure according to the four general principles of Chinese Pharmacopoeia(2020 edition).The water content was determined by drying method,the total ash and acid insoluble ash were determined by Muffle furnace,the contents of extractives(water soluble and ethanol soluble)were determined by soaking method and heating reflux method,and the contents of heavy metals Pb,Cu,As,Cd and Hg were determined by inductively coupled plasma emission spectrometer ICP-OES.The results showed that the microscopic characteristics of stems and leaves samples from different places are basically the same.Thin layer chromatography(TLC)presented good reproducibility and specificity.Water content was 6.83%-9.16%,total ash content was 3.74%-5.90%,acid insoluble ash content was 0.49%-0.92%.The content of water soluble extract(cold leaching)was 3.01%-10.78%,water soluble extract(hot leaching)was 5.88%-15.03%,ethanol extract(cold leaching)was 2.38%-4.22%,ethanol extract(hot leaching)was 3.29%-6.36%.Pb content ranged from 0.185 to 2.863 mg/kg,Cu content ranged from 1.840 to 6.601 mg/kg,As content ranged from undetected to 0.028 mg/kg,Cd content ranged from 0.016 to 0.086 mg/kg,Hg content ranges from undetected to 0.019 mg/kg.This test method is simple,feasible and reliable,and can provide reference for the establishment of the qualitative quality standard of the stems and leaves of Millettia speciosa Champ..
