To optimize and scientific evaluate the extraction process of modified Gegen Qinlian decoction,so as to lay the foundation for its better application in clinical dialectics as well as the development of new veterinary drugs.Box-Behnken design-response surface method was used to investigate three influencing factors, the times of extraction, the decoction time, and water addition, using the content of puerarin as the evaluation index.17 batches of samples were quantitatively analyzed under the response surface conditions, and the degree of influence of the factors on the extraction rate of puerarin was obtained.Through Box-Behnken center set and design,a mathematical model was established to analyze regression model variance and response surface curve. The results showed that the optimized modified Gegen Qinlian decoction had the best extraction process of three times, extraction time of 2 h,and 9 times the amount of water addition, and the obtained mean value of puerarin content was 0.810 8 mg/mL which was close to the predicted value of 0.811 0 mg/mL.The derived optimal extraction process of modified Gegen Qinlian decoction is characterized by high dissolution rate of active ingredients, stable and feasible, which can be used as a theoretical basis for the further study of its compound formulations.
