Comparison of potentiometric titration and high performance liquid chromatography for the determination of barbital content in veterinary compound preparations
The objective was to analyze and compare the differences between potentiometric titration and high performance liquid chromatography in the determination of barbiturate content,so as to provide reference for practical detection work.A 916 Ti-Touch automatic potentiometric titrator was used to perform potentiometric titration with silver nitrate titrant.High performance liquid chromatography was carried out by using Xbridge C18(150.0 mm×4.6 mm,5 μm)as the chromatographic column;the mobile phase was acetonitrile-0.05 mol/L potassium dihydrogen phosphate solution(volume ratio 25∶75,adjusted to pH 8.2 with triethylamine);the detection wavelength was 240 nm;column temperature was 30℃with flow rate of 1.0 mL/min and injection volume of 10 μL.The result showed that the recovery rate of the potentiometric titration method was 100.2%-101.2%with RSD of 0.1%;the recovery rate of high performance liquid chromatography was 99.0%-101.6%with RSD of 0.1%-0.2%.The barbiturate content in 12 batches of animal compound aminopyrine injection and Antongding injection from different manufacturers was determined by potentiometric titration and high performance liquid chromatography,respectively.The results of the two methods were not significantly different(P>0.05).High performance liquid chromatography is time-consuming and costly,making it more suitable for the simultaneous determination of multiple components in compound preparations.The potentiometric titration method is simple and fast to operate,with high accuracy and good precision,and is more suitable for the determination of the content of a single component of barbiturate.
