Pachymaran is the main active component of Pachymaran powder to regulate immune function.The content of pachymaran in Pachymaran powder was determined with phenol-sulfuric acid method by spectrophotometry.The optimum test conditions were selected from six aspects:solvent,wavelength,reaction temperature,reaction time,addition amount of 5%phenol solution and addition amount of sulfuric acid.The linear relationship,precision,stability,repeatability and sample recovery method were further verified.The results showed that the optimal test conditions for the determination of Pachymaria polysaccharide were 50%sulfuric acid solution in solvent,the determination wavelength was 490 nm,the reaction time was 40 minutes at 25℃ water bath,the addition amount of 5%phenol was 1.8 mL,and the addition amount of sulfuric acid was 6.0 mL.D-anhydrous glucose had good linear relationship in the range of 10.19 μg/mL to 61.14 μg/mL(R2=0.999 4,n=6),and the average recovery was 99.31%.The method has the advantages of high sensitivity,precision,stability,good repeatability,accuracy and reliability,and can be used as a control method for polysaccharide content of Pachymaran powder.
