Quality control of Jigen pieces using HPLC characteristic chromatogram combined with chemical pattern recognition method
The objective was to establish a quality control method for Jigen pieces based on characteristic chromatogram and component determination.Agilent ZORBAX Extend-C18(4.6 mm×250.0 mm,5 μm)column was used for gradient elution with methanol-0.1%phosphoric acid solution,wavelength of 330 nm,flow rate of 1.0 mL/min,column temperature of 30℃,and injection volume of 5 μL.The characteristic chromatogram was established by the Similarity Evaluation System of Traditional Chinese Medicine Fingerprint,and the main differential components of Jigen pieces between different batches were analyzed by cluster analysis,principal component analysis and orthogonal partial least squares discrimination,and the content of 3,6'-diglucoylsucrose was determined.Based on the characteristics of 12 batches of Jigen pieces in Yunnan producing area,four common peaks were determined,and one component was identified as 3,6'-diglucoylsucrose,and the similarity of the characteristic maps was greater than 0.860.Cluster analysis divided the samples into two categories:principal component analysis(PCA)was used to screen out two principal components with eigenvalues greater than 1,and orthogonal partial least squares discriminant analysis(VIP>1)was used to screen out two landmark components with mass differences,such as 3,6'-diglucoylsucrose.The average recovery rate was 98.683%and the RSD was less than 3.00%,the results showed that there were differences in the content of 3,6'-diglucoylsucrose in 15 batches of Jigen pieces from five producing areas.Among them,the content of 3,6'-diglucoylsucrose in Simao area was the highest.The characteristic map and content determination method of Jigen pieces established in this study are accurate,stable and reliable,which can provide a reference for controlling and evaluating the quality of Jigen pieces.
