Tubiechong is a dried female of Eupolyphaga sinensis Walker or Steleophaga plancyi Boleny,while the male worm can not be used as medicine.In order to improve the safety and effectiveness of drugs,it should to accurately and effectively distinguish the male and female of Tubiechong.High performance liquid chromatography(HPLC)was performed on Zafex SupperfexAQ-C18(4.6 mm×250.0 mm,5 μm)column.Mobile phase A was methanol,and mobile phase B was 0.1%glacial acetic acid solution in gradient elution of 60 min.The column temperature was 30℃.The detection wavelength was 248 nm and sample volume was 10 μL.The results showed that the chromatographic peaks of 8 characteristic peaks corresponding to the control materials of 28 batches of Tubiechong(female)were found.4 chemical components were identified by the control materials,namely uracil,hypoxanthine,xanthine and guanosine.And it did not match the fingerprint of the male.This method can effectively distinguish male and female of Tubiechong,and can effectively avoid adulteration,which can provide reference method for quality control of Tubiechong.
