Virulence and infectious characteristics of a Spodoptera frugiperda multiple nucleopolyhedrovirus to fall armyworm Spodoptera frugiperda
To investigate the virulence of a Spodoptera frugiperda multiple nucleopolyhedrovirus (SfM-NPV) to fall armyworm Spodoptera frugiperda and its infectious characteristics in vivo and in vitro,bio-assays were conducted to determine the virulence of SfMNPV in the laboratory. Histopathological analysis of S. frugiperda infected by SfMNPV was carried out by using hematoxylin-eosin (HE) stain-ing. Real-time fluorescence quantitative PCR was used to analyze the replication characteristics of SfM-NPV in S. frugiperda larvae and Sf9 cells. The results showed that the LC50 and LT50 of SfMNPV to the second-instar larvae of S. frugiperda were 7.39×105 OBs/mL and 3.82 d,respectively. The epidermis,trachea,fat body,and midgut tissues of the larvae could be infected by SfMNPV,while muscle tissue was not significantly infected. The genome copies of SfMNPV in S. frugiperda larvae reached a maxi-mum of 3.91×106 copies/ng at 96 h post-infection. The genome copies of SfMNPV in the Sf9 cell cul-ture medium supernatant peaked at 2.08×107 copies/mL at 96 h post-infection.
