基于数据挖掘和网络药理学分析治疗失眠的用药规律及其药理机制

目的 利用数据挖掘分析治疗失眠的用药规律,并应用网络药理学初步分析药物的作用机制.方法 收集 2022 年 8月至 2023 年 9 月梁晖治疗失眠的处方,应用Excel软件和中医传承辅助平台等工具构建数据库,对药物进行频次、药性、药物关联规则及聚类分析等,检索核心药物成分及疾病靶点,对其治疗失眠作用靶点进行基因本体和京都基因与基因组百科全书分析.结果 共收集失眠处方 300 首,涉及药物 96 味;药性以寒性为主,药味多偏苦味;其配伍核心组合包括柴胡、茯苓、白术、白芍、郁金、栀子、知母、茯神、养心草.治疗失眠药物的关键靶点包括白蛋白、丝氨酸苏氨酸蛋白激酶 1、表皮生长因子受体、B淋巴细胞瘤-2、雌激素受体 1、胱天蛋白酶-3 等;主要富集通路包括癌症通路、脂质和动脉粥样硬化通路、磷脂酰肌醇 3 激酶(phosphoinositide 3-kinase,PI3K)/蛋白激酶B(protein kinase B,PKB,又称Akt)信号通路、卡波西肉瘤相关疱疹病毒感染通路、缺氧诱导因子-1(hypoxia-inducible factor-1,HIF-1)信号通路等;主要生物学过程包括细胞凋亡、对尼古丁的反应、丝裂原活化蛋白激酶级联反应等.结论 梁晖治疗失眠以标本同治为原则,重在清利三焦,调肝健脾,畅达气机,佐以养心安神,形成以柴胡、茯苓、白术、郁金、栀子、知母、茯神、养心草于一体的核心处方,其作用通路与PI3K/Akt、HIF-1 等相关.
Analysis of medication patterns and pharmacological mechanisms for treating insomnia based on data mining and network pharmacology
Objective Using data mining to analyze the medication patterns for treating insomnia,and applying network pharmacology to preliminarily analyze the mechanisms of drugs.Methods The prescriptions for the treatment of insomnia by Liang Hui from August 2022 to September 2023 were collected.A database was constructed using tools such as Excel software and the Traditional Chinese Medicine Inheritance Auxiliary Platform.The drugs were analyzed for frequencies,natures,drug associations,and clustering were analyzed.Core drug components and disease targets were retrieved,and the target genes for the treatment of insomnia were subjected to gene ontology and Kyoto Encyclopedia of Genes and Genomes analysises.Results A total of 300 insomnia-related prescriptions were collected,involving 96 flavors of drugs.The drug nature was mainly cold,and the drug flavor was mainly bitter.The core herbal combinations included Radix Bupleuri,Poria,Atractylodis Macrocephala,Radix Paeoniae Alba,Radix Curcumae,Fructus Gardeniae,Anemarrhenae,Poria cum Radix Pini,Sedum aizoon L.Critical drug targets for the treatment of insomnia encompassed albumin,serine/threonine kinase 1,epidermal growth factor receptor,B-cell lymphoma-2,estrogen receptor 1,cysteinyl aspartate specific proteinase-3,among others;The main enriched pathways included cancer pathway,lipid and atherosclerosis pathway,phosphoinositide 3-kinase(PI3K)/protein kinase B(PKB,also known as Akt)signaling pathway,Kaposi sarcoma-associated herpesvirus infection pathway,hypoxia-inducible factor-1(HIF-1)signaling pathway,etc.;The main biological processes included cell apoptosis,response to nicotine,mitogen-activated protein kinase cascade reactions,etc..Conclusion Liang Hui's treatment of insomnia follows the principle of treating symptoms with the same root cause,focusing on Qingli Sanjiao,regulating liver and strengthening spleen,smoothing the qi and organs,supplementes by nourishing the heart and calming the nerves,forming a core prescription integrating Radix Bupleuri,Poria,Rhizoma Atractylodis Macrocephalae,Radix Paeoniae Alba,Radix Curcumae,Fructus Gardeniae,Rhizoma Anemarrhenae,Poria cocos,Sedum aizoon L.Its pathways are related with PI3K/AKT,HIF-1 and so on.
