厌食康颗粒干预特制饲料叠加夹尾致小儿厌食症大鼠模型的药效学研究

目的 探究厌食康颗粒对特制饲料叠加夹尾致小儿厌食症大鼠模型的治疗作用.方法 采用特制饲料叠加夹尾幼龄大鼠的方法建立小儿厌食症大鼠模型.大鼠随机分为对照组、模型组及厌食康颗粒低、中、高剂量(2.8、5.6、11.1 g·kg-1)组和儿滞灵冲剂(10.1 g·kg-1)组.监测各组大鼠的情绪行为状态、体质量、摄食量;采用MP150多导生理记录仪系统检测十二指肠生物电信号的频率和振幅;酶联免疫吸附测定法(ELISA)检测血清中瘦素(LEP)、生长抑素(SST)、胃动素(MTL)及胃泌素(GAS)水平;16SRNA高通量测序技术分析肠道菌群结构.结果 与模型组相比,厌食康颗粒给药后大鼠摄食量和体质量显著增长(P<0.05、0.001);十二指肠生物电信号振幅和频率显著升高(P<0.05、0.01);血清LEP和SST水平显著降低(P<0.05、0.01、0.001)、MTL水平升高(P<0.05、0.01);肠道菌群Firmicutes丰度显著降低(P<0.05).结论 厌食康颗粒对特制饲料叠加夹尾致小儿厌食症大鼠模型具有明显的治疗作用,其作用机制与调节胃肠激素、改善肠道微生物环境有关.
Pharmacodynamic study on intervention of Yanshikang Granules in rat model of pediatric anorexia induced by special forage and tail clipping
Objective To investigate the therapeutic effect of Yanshikang Granules in a rat model of pediatric anorexia induced by a special forage and tail clipping.Methods A rat model of pediatric anorexia nervosa was established using a special chow stacked on a tail-clamped young rat.The rats were randomly divided into six groups,including control group,model group,Yanshikang Granules low,medium,high dose(2.8,5.6,11.1 g·kg-1)groups,and Erzhiling Chongji(10.1 g·kg-1)group.The general status,body weight and food intake of each group of rats were monitored,and the frequency and amplitude of the duodenal bioelectrical signal was detected by MP 150 system,serum levels of leptin(LEP),somatostatin(SST),motilin(MTL),and gastrin(GAS)were measured by ELISA method,the structure of gut microbial community were measured by 16S RNA high-throughput sequencing technology.Results Compared with the model group,after treatment with Yanshikang Granules,food intake and body weight were significantly improved(P<0.05,0.01),amplitude and frequency of duodenal bioelectrical signal were significantly improved(P<0.05,0.01),serum LEP and SST were significantly reduced(P<0.05,0.01,0.001),serum MTL were significantly improved(P<0.05,0.01),the abundance of Blautia were significantly reduced(P<0.05).Conclusion Yanshikang Granules can treat the anorexia situation in a rat model of anorexia caused by the combination of special forage and tail clamping.The mechanism is related to regulating the secretion of gastrointestinal hormone and improving the gut microbial environment.
