牛磺酸是一种带有氨基的磺酸,是人体所需营养成分之一。目前,牛磺酸广泛应用于乳制品、饮料、复合味精、豆制品、滋补品、营养液,还可用于水产、动物养殖饲料添加剂,洗涤剂中间体和化学剂等。它也是鱼虾类食品风味形成的呈味氨基酸。半胱氨酸双加氧酶是牛磺酸生物合成的两个限速酶之一,在牛磺酸的合成中起关键作用。本研究利用race PCR技术克隆到了凡纳滨对虾(Litopenaeus vannamei)的半胱氨酸双加氧酶(L. vannamei cysteine dioxygenase, LvCDO)基因的全长序列,对其编码的蛋白的特性进行了分析。同时构建原核表达载体对其进行表达,获得可溶表达的LvCDO,并利用GST琼脂糖凝胶对其进行纯化,获得较纯的重组GST-LvCDO。另外还发现在盐度为15‰的养殖水体中, LvCDO在凡纳滨对虾的血淋巴细胞和肝胰腺细胞中有较高的转录水平,表明其调节渗透压方面具有一定功能。
Taurine is one of the necessary nutrients for animal. At Nowadays, taurine is widely used in dairy products, beverage, composite monosodium glutamate, bean products, tonic, nutrient solution, animal/aquaculture feed additive, animal, intermediates and chemical agents. It is also the delicious amino acid of fish and shrimp. Cysteine dioxygenase is one of the rate limiting enzyme in the biosynthesis of taurine. In this study, full-length of Litopenaeus vannamei cysteine dioxygenase gene (LvCDO) was obtained by race PCR. And then the characteristics of the LvCDO were analyzed. At the same time, prokaryotic expression vector was constructed to get soluble expression LvCDO, and it was purified by GST agarose. We also found that LvCDO was upregulated upon osmotic stress in haemocytes and hepatopancreas of L. vannamei.
