Isolation of endophytic fungi from the leaves of Cichorium intybus and analysis on their antifungal activity against phytopathogens
Endophytic fungi were isolated from the leaves of Cichorium intybus Linn.by using the tissue separation method,endophytic fungi with antagonistic activity against nine common phytopathogens were screened by using the plate confrontation method,the endophytic fungi were identified by using a combination of morphology and molecular biology,and the antifungal effects of the fermentation products of the active endophytic fungi were assayed.The results show that a total of 24 endophytic fungi are isolated from the leaves of C.intybus,one endophytic fungal isolate CIY4 that shows good antagonistic activity against all nine phytopathogens is screened out,and it is identified as Alternaria destruens belonging to the genus Alternaria Nees.The inhibition rate of 500 μg·mL-1 fermentation products of isolate CIY4 against Rhizoctonia solani J.G.Kühn reaches 86.31%,followed by Sclerotinia sclerotiorum Lib.(85.45%),Colletotrichum acutatum J.H.Simmonds(77.87%),and Colletotrichum gloeosporioides Penz.(71.12%),while the inhibition rates against the other phytopathogens are lower than 70%.This study isolates and screens the endophytic fungal isolate CIY4 from the leaves of C.intybus for the first time,and this Alternaria fungus exhibits a broad-spectrum inhibitory effect against common phytopathogens.
