Study on Total Phenolic Content and Correlation between PPO Activity and Browning of Corydalis saxicola Bunting Callus
With non-browned callus of Corydalis saxicola as materials,the effects of different concentrations of ascorbic acid on browning of Corydalis saxicola callus,total phenolic content and PPO activity were studied by adding them to culture medium in this experiment.The results showed that the browning rates of Corydalis saxicola bunting callus under different ascorbic acid concentrations were lower than that of the control on the 20th day.While the activities of PPO of all the treatments were higher than that of the control,the activity of PPO with concentration of 175mg/L ascorbic acid was the highest with a value of 14.1400U/(g · min),72.3% higher than the control.The total phenolic contents of all the treatments were lower than that of the control,and the total phenolic content with concentration of 175mg/L ascorbic acid was the lowest with a value of 0.4633μg/mL,88.3% lower than the control.The correlation analysis showed that the browning rate of callus was positively correlated with the content of total phenols,and negatively correlated with the activity of PPO.The activity of PPO was much negatively correlated with the total phenol content in the callus.
