Exploring the medication pattern and mechanism of action in liver depression and spleen deficiency type liver cancer based on data mining and network pharmacology
Objective:Unearthing the medicinal formulas and pattern of treatment for liver cancer of liver depression and spleen deficiency syndrome in traditional Chinese medicine,analyzing the drug targets and disease pathways associated with prescriptions,and exploring the potential mechanisms of core prescriptions.Methods:The clinical literature of TCM on liver cancer in CNKI,VIP and Wanfang were searched.The frequency and property of TCM were statistically analyzed by Microsoft Excel2021,and the association rules were analyzed by SPSS Modeler 18.0 software Apriori algorithm.SPSS 25.0 software was used to analyze the rules of prescription and medication.The TCMSP platform was used to search the active components and targets of the core drugs,and the liver cancer target genes were obtained in the human gene database GeneCards database.The core drug pair and liver cancer target genes were simultaneously imported into Venny 2.1.0 platform to obtain the drug-disease intersection targets.Cytoscape3.9.0 was used to construct the drug-disease-target gene network,and the metascape platform was used for gene ontology(GO)function and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis.Results:A total of 45 articles and 83 prescriptions were screened out.The top five high-frequency drugs were Radix bupleurum,Atractylodes atractylodes,Poria cocos,Licorice and Radix paeoniae.The main medicinal properties were warm and cold,the main medicinal flavors were spicy,sweet and bitter,and the main meridians were liver,spleen,lung and stomach meridians.A total of 20 groups of related drugs were obtained,and 4 core drug prescriptions were clustered.A total of 112 core drug active ingredients,209 drug targets,16458 disease targets,and 204 disease-drug intersection targets were obtained.GO functional enrichment analysis identified 1840 biological processes,113 cellular components and 215 molecular functions,and KEGG pathway enrichment analysis identified 209 signaling pathways.Conclusion:The traditional Chinese medicine for the treatment of liver stagnation and spleen deficiency liver cancer mainly regulates the key targets such as Akt1,interleukin-6,and interleukin-1b recombinant protein.
