Artesunate down-regulates lncRNA HOTAIR expression and inhibits proliferation and migration of hepatocellular carcinoma cells
Objective:To investigate the effect and mechanism of Artesunate on the proliferation and migration of human hepatocellular carcinoma cells by inhibiting the expression of lncRNA HOTAIR.Methods:The effects of different concentrations of artesunate on the proliferation and migration of Huh7 and HepG2 cells were detected by MTT assay and Transwell chamber method.The effect of ART on HOTAIR expression in Huh7 cells was detected by qRT-PCR method.Huh7 cells were transfected with HOTAIR overexpression plasmid,and the transfection efficiency was verified by qRT-PCR.Normal Huh7 cells and Huh7+HOTAIR-OE cells were treated with different concentrations of ART,and then MTT assay and Transell chamber method were used to detect the changes of proliferation and migration ability of ART.To verify the effect of HOTAIR overexpression on ART on hepatoma,and to further understand whether ART can inhibit hepatoma cells by regulating HOTAIR.Results:ART inhibited the proliferation and migration of human hepatoma Huh7 and HepG2 cells in a dose-dependent manner.ART could inhibit the expression of HOTAIR in Huh7 cells.Overexpression of HOTAIR attenuated the inhibitory effect of ART on proliferation and migration of Huh7 cells.Conclusion:ART may inhibit the proliferation and migration of hepatocellular carcinoma cells through or partly down-regulating the expression of lncRNA HOTAIR.
