Bioinformatics analysis of genes associated with Helicobacter pylori infection and non-alcoholic fatty liver disease
Objective:Bioinformatics analysis of Helicobacter pylori and non-alcoholic fatty liver disease gene chips was used to obtain common differentially expressed genes(DEGs)and predict the corresponding therapeutic herbs and effective active ingredients. Methods:The GSE5081 and GSE63067 datasets were screened in the GEO database, and the DEGs were screened and enriched by R software. With the help of STRING database and Cytoscape,protein-protein interaction network was constructed to screen out key genes. Then, the traditional Chinese medicine and active ingredients corresponding to the key genes were analyzed in the Coremine Medical and CTD databases. Results:A total of 14 common DEGs were screened from the two datasets, and the enrichment analysis was mainly related to inflammatory cells, glycolipid metabolism, and signaling pathways such as IL-17,NF-κB,and TNF.The targeted Chinese medicines are mainly of two types:expelling dampness and resolving phlegm, activating blood circulation and removing blood stasis, and among the active ingredients of the Chinese medicines, resveratrol, curcumin, sulforaphane, folic acid, and genistein appeared with the highest frequency. Conclusion:Bioinformatics screening identified common DEGs between Hp infection and NAFLD,and obtained eight key genes through protein-protein interactions network, which provided a new idea for the mechanism study of the relationship between the two occurrences and development as well as disease control.
