Effects of Icariin on Memory Function and Expressions of Glial Fibrillary Acidic Protein,Tumor Necrosis Factor-α and Interleukin 6 in Hippocampus of Rats with Alzheimer's Disease
Objective To investigate the influence of icariin (ICA) on memory of rat model with Alzheimer's disease (AD) induced by lipopolysaccharides. Methods After successfully trained in the 8 - arm maze, 36 rats were allocated into 4 groups:Control group, model group, low - dose ICA group, and high - dose ICA group. Then the rat models with AD were induced by injection of lipopolysaccharides (LPS) into the lateral cerebral ventricle. The spatial memory dysfunction of the rats was detected by 8 - arm maze test. The expressions of glial fibrillary acidic protein (GFAP), tumor necrosis factor alpha (TNF - α), interleukin 6 (IL - 6) in hippocampus of the rats were detected by immunohistochemistry (SP method). Results After administration, the memory impairment of rats injected LPS was markedly inhibited in high - dose ICA group. The total error(TE), reference memory error(RME) and working memory er-ror(WME)in high - dose ICA group were significantly decreased compared with those in model group (P<0. 01). There was no significant difference in TE, RME, WME in low - dose ICA group, compared with those in model group (P>0. 05). The expressions of GFAP, TNF- α and IL - 6 in high - dose ICA group were significantly lower than those in model group (P<0. 01). There was no difference in expressions in low - dose ICA group compared to those in model group (P>0. 05). Conclusion ICA could ameliorate the ability of study and memory of the rat model with AD induced by LPS. It might inhibit the hyperplasia of astrocytes and decrease the expressions of GFAP, TNF - α and IL - 6 in hippocampus.
