麝香通心滴丸预处理对大鼠心肌缺血再灌注损伤的保护作用机制

目的:观察麝香通心滴丸预处理对大鼠心肌缺血再灌注(I/R)损伤后心肌细胞和血管的保护作用及潜在机制。方法:将 60 只雄性 SD大鼠随机分为假手术组(Sham组)、I/R组、麝香通心滴丸组(SXTX组)和阳性组,每组 15只。SXTX组以 18。90 mg/(kg·d)的浓度灌胃;阳性组以尼可地尔 1。35 mg/(kg·d)的浓度灌胃;Sham组和 I/R组给予等体积去离子水灌胃。各组均灌胃 1 周后,行冠状动脉前降支结扎-松解手术,除 Sham组外其余各组大鼠结扎左冠状动脉前降支 30 min后,再灌注 120 min,构建心肌 I/R模型。通过心肌苏木精-伊红(HE)染色、心肌 Masson染色观察大鼠心肌组织及血管改变;电镜观察心肌细胞和血管的超微结构改变;免疫组织化学法观察血管相关蛋白低氧诱导因子-1α(HIF-1α)和血管内皮生长因子(VEGF)表达情况。结果:与 Sham组比较,I/R组大鼠心肌细胞坏死和血管损伤明显,心肌组织中 HIF-1α表达增加;与 I/R 组相比,SXTX组心肌细胞和血管病理结构损伤减轻,缺血区心肌组织 VEGF含量明显增加。结论:麝香通心滴丸预处理可保护大鼠心肌细胞和血管 I/R损伤,可能与上调 VEGF蛋白表达、促进缺血区微血管新生有关。
Protective Effect of Shexiang Tongxin Dropping Pill Pretreatment on Myocardial Ischemia-reperfusion Injury in Rats
Objective:To observe the protective effect of Shexiang Tongxin Dropping Pills on myocardial cells and blood vessels after myocardial ischemia-reperfusion(I/R)injury in rats.Methods:Sixty male SD rats were randomly divided into sham operation group(Sham group),I/R group,Shexiang Tongxin Dropping Pills group(SXTX group),and positive control group,with 15 rats in each group.The rats in SXTX group were given SXTX 18.90 mg/(kg·d)by gavage,and the rats in positive control group were given nicorandil 1.35 mg/(kg·d)by gavage.The rat in Sham group and I/R group were given the same volume of deionized water by gavage.After one week of gavage,the rats left anterior descending coronary artery was ligated for 30 min and then reperfused for 120 min to build the myocardial I/R model except for the Sham group.The changes of myocardial tissue and blood vessels in rats were observed by myocardial hematoxylin-eosin(HE)staining and myocardial Masson staining.The ultrastructural changes of cardiomyocytes and blood vessels were observed by electron microscope.The expressions of hypoxia-inducible factor-1α(HIF-1α)and vascular endothelial growth factor(VEGF)were observed by immunohistochemistry.Results:Compared with Sham group,the myocardial cell necrosis and vascular injury of the rats in I/R group were obvious,and the expression of HIF-1αin the myocardial tissue increased.Compared with I/R group,the myocardial cell and vascular pathological structural damage in SXTX group alleviated,and the protein expressions of VEGF in the ischemic area significantly increased.Conclusion:Shexiang Tongxin Dropping Pills pretreatment can protect myocardial cells and vascular I/R injury in rats,which may be related to the up-regulation expression of VEGF protein and the promotion of microangiogenesis in ischemic areas.
