Objective:To investigate the effect of Tianxiangdan on purinergic ligand-gated ion channel 7(P2X7)and nucleotide-binding oligomerization domain-like receptor protein 3(NLRP3)in atherosclerotic mice.Methods:Thirty 8-week-old SPF male ApoE-/-mice were selected and fed with high-fat diet for 12 weeks.After successful modeling,they were randomly divided into model group,statin group,and Tianxiangdan group,with 10 mice in each group.Ten C57BL/6J mice were fed with normal diet as blank control group.The levels of interleukin(IL)-18,IL-1β,and adenosine triphosphate(ATP)in serum were detected by enzyme-linked immunosorbent assay(ELISA).The expression of P2X7 and NLRP3 in tissues was detected by immunohistochemistry.Results:ELISA results displayed that compared with the blank group,the levels of IL-18,IL-1β,and ATP in model group increased(P<0.05).Compared with the model group,the levels of IL-18,IL-1β,and ATP in statin group and Tianxiangdan group decreased(P<0.05).The results of immunohistochemistry showed that the expression of P2X7 and NLRP3 in model group was significantly higher than that in blank group(P<0.05).The expression of P2X7 and NLRP3 in the statin group and Tianxiangdan group decreased compared with the model group(P<0.05).Conclusion:The improvement of atherosclerosis by Tianxiangdan is related to regulating potassium homeostasis and inhibiting the activation of inflammasome.
