天香丹抑制P2X7/NLRP3表达改善动脉粥样硬化的作用机制研究

目的:探讨天香丹对动脉粥样硬化小鼠嘌呤能配体门控离子通道 7(P2X7)、核苷酸结合寡聚化结构域样受体蛋白 3(NLRP3)的影响。方法:选取 8周龄无特定病原体(SPF)级雄性载脂蛋白 E基因敲除(ApoE-/-)小鼠 30 只,予以高脂饲料喂养 12 周,造模成功后随机分为模型组、阿托伐他汀组、天香丹组,每组 10只;另选取 C57BL/6J小鼠 10只予普通饲料喂养,设为空白对照组。酶联免疫吸附法(ELISA)检测血清白细胞介素(IL)-18、IL-1β、腺嘌呤核苷三磷酸(ATP)的水平;免疫组化检测组织中 P2X7、NLRP3 的表达。结果:ELISA结果显示:与空白对照组相比,模型组 IL-18、IL-1β、ATP水平升高(P<0。05);与模型组相比,阿托伐他汀组、天香丹组IL-18、IL-1β、ATP水平降低(P<0。05)。免疫组化结果显示:与空白对照组相比,模型组 P2X7、NLRP3 表达均明显升高(P<0。05);与模型组相比,阿托伐他汀组、天香丹组 P2X7、NLRP3表达均明显降低(P<0。05)。结论:天香丹改善动脉粥样硬化与调节钾稳态抑制炎症小体的激活有关。
The Effect of Tianxiangdan on Atherosclerosis by Inhibiting P2X7/NLRP3 Expression
Objective:To investigate the effect of Tianxiangdan on purinergic ligand-gated ion channel 7(P2X7)and nucleotide-binding oligomerization domain-like receptor protein 3(NLRP3)in atherosclerotic mice.Methods:Thirty 8-week-old SPF male ApoE-/-mice were selected and fed with high-fat diet for 12 weeks.After successful modeling,they were randomly divided into model group,statin group,and Tianxiangdan group,with 10 mice in each group.Ten C57BL/6J mice were fed with normal diet as blank control group.The levels of interleukin(IL)-18,IL-1β,and adenosine triphosphate(ATP)in serum were detected by enzyme-linked immunosorbent assay(ELISA).The expression of P2X7 and NLRP3 in tissues was detected by immunohistochemistry.Results:ELISA results displayed that compared with the blank group,the levels of IL-18,IL-1β,and ATP in model group increased(P<0.05).Compared with the model group,the levels of IL-18,IL-1β,and ATP in statin group and Tianxiangdan group decreased(P<0.05).The results of immunohistochemistry showed that the expression of P2X7 and NLRP3 in model group was significantly higher than that in blank group(P<0.05).The expression of P2X7 and NLRP3 in the statin group and Tianxiangdan group decreased compared with the model group(P<0.05).Conclusion:The improvement of atherosclerosis by Tianxiangdan is related to regulating potassium homeostasis and inhibiting the activation of inflammasome.
