Effect of Tongluo Xifeng Decoction on Myocardial Injury in Spontaneously Hypertensive Rats by Regulating Mst1/Sirt3 Signaling Pathway
Objective:To clarify the therapeutic effect on hypertensive myocardial injury in rats.Methods:Six Sprague-Dawley rats were used as blank group,and 12 spontaneously hypertensive rats were randomly divided into model group and Tongluo Xifeng decoction group.Rats in blank group and model group were given deionized water intragastrically,and Tongluo Xifeng decoction group was given TCM formula granule Tongluo Xifeng decoction suspension intragastrically for 8 weeks.The contents of autophagy receptor protein 1(SQSTM1/P62),phosphorylated mammalian aseptic 20-like kinase 1(p-Mst1)and silence-information regulator 3(Sirt3)protein in cardiomyocytes were detected by Western Blot.The changes of myocardial tissue structure in rats were observed by hematoxylin-eosin(HE)staining to evaluate the effect of Tongluo Xifeng decoction on hypertensive myocardial injury.Results:Compared with model group,Tongluo Xifeng decoction group could significantly improve systolic and diastolic blood pressure(P<0.01).After treatment with Tongluo Xifeng decoction,the arrangement and necrotic state of rat cardiomyocytes significantly improved compared with that of model group.The protein contents of P-MST1 and P62 in myocardial tissue of Tongluo Xifeng decoction group significantly decreased compared with that of model group(P<0.01),while the protein content of Sirt3 significantly increased(P<0.01).Conclusion:Tongluo Xifeng decoction can mediate Mst1/Sirt3 signaling pathway to adjust the autophagy level of myocardial tissue cells and protect cardiac function.
